EZH2 Is Required for Human Hair Follicle Growth and Epidermal Differentiation

The study demonstrated that Enhancer of zeste homolog 2 (Ezh2) was crucial for human hair follicle growth and epidermal differentiation. Ezh2 was predominantly expressed in epithelial cells of human skin, particularly in differentiating cells of the suprabasal layer and anagen hair follicles. Knockdown of EZH2 and inhibition with small molecules led to increased expression of differentiation markers and a dose-dependent retardation of hair growth, indicating decreased proliferation and increased apoptosis in the outer root sheath. These findings suggested that Ezh2 played a vital role in regulating the proliferation and differentiation of human epithelial progenitor cells.
