TLDR Finasteride may cause sexual and psychological side effects by affecting an enzyme related to epinephrine.
The study explored the potential off-target effects of finasteride, a drug commonly used for androgen-dependent conditions, by identifying its interaction with phenylethanolamine N-methyltransferase (PNMT), an enzyme involved in epinephrine formation. Using SPILLO-PBSS software, docking, molecular dynamics analysis, and in vitro assays, researchers confirmed finasteride's inhibitory effect on PNMT. This inhibition was further validated in an in vivo rat model. The findings suggested that finasteride's interaction with PNMT could contribute to the sexual and psychological side effects reported by users, indicating that mechanisms beyond 5-alpha reductase inhibition might be involved in these adverse effects.
