CUBIC Protocol Visualizes Protein Expression at Single Cell Resolution in Whole Mount Skin Preparations

    TLDR The CUBIC protocol allows detailed 3D visualization of proteins in mouse skin biopsies.
    The CUBIC protocol provided a method to clarify full thickness mouse skin biopsies and visualize protein expression at single cell resolution in 3D. It enabled the assessment of K14 protein expression, Ki67 labeled proliferating cells, Nile Red labeled sebocytes, and DAPI nuclear labeling. This method allowed for accurate evaluation of skin anatomy, pathology, and abnormal epidermal phenotypes in genetically modified mouse lines, making it the best available technique for investigating molecular and cellular interactions in full thickness skin biopsies at the time.
