The study investigated the penetration pathways of 15 nm iron oxide nanoparticles (INPs) through the skin of C57BL/6 mice, focusing on the stratum corneum and hair follicles. Using transmission electron microscopy (TEM), researchers found that INPs primarily permeated the upper stratum corneum, hair follicles, and sebocytes via the pilosebaceous duct. The amount of INPs increased in sebocytes with longer application times, and INPs were also observed in the upper hair bulb through the inner root sheath and hair cuticle interface. However, INPs did not permeate the dermis outside of hair follicles and sebocytes. The findings provided valuable data for transdermal drug delivery research.
