Formulation and Evaluation of Self-Nanoemulsifying Drug Delivery System for Improved Oral Delivery of Exemestane Hydrochloride

    The study focuses on improving the solubility and oral delivery of Exemestane HCl (EXM), a steroidal aromatase inhibitor used in breast cancer therapy, by developing a self-nanoemulsifying drug delivery system (SNEDDS). Using a water titration approach, the researchers selected Caprol Microexpress and Labrafac as the oil phase, Tween 80 as the surfactant, and Triacetin as the co-surfactant. Various surfactant:co-surfactant ratios were tested, with Tween 80:Triacetin (1:2) and (1:3) showing the largest nanoemulsion areas. Among 10 formulations, F3, F7, and F8 had the smallest particle sizes and rapid self-emulsification times, with F7 maintaining optical clarity without precipitation for 24 hours. The SNEDDS formulation demonstrated potential for enhanced dissolution and bioavailability of EXM.
