    This study explored a needle-free transcutaneous vaccination strategy using cyanoacrylate skin surface stripping (CSSS) combined with particle-based antigen delivery to target the HIV-1 p24 protein to skin antigen presenting cells (APC). The CSSS method pre-activated skin APC and opened hair follicles, allowing protein-loaded particles to accumulate and deliver the antigen to perifollicular APC. Poly-lactic acid (PLA) and polystyrene (PS) particles effectively targeted the HIV-1 p24 protein to hair follicles, with some particles translocating to the epidermis and being internalized by skin cells. The p24 protein was detected in skin cells, particularly Langerhans cells and dermal dendritic cells, and the approach led to the activation and maturation of Langerhans cells. The study concluded that this method was a feasible and effective approach for transcutaneous vaccination.
