Faculty Opinions Recommendation of Identification of Drug-Specific Public TCR Driving Severe Cutaneous Adverse Reactions

TLDR Researchers found a specific T cell receptor linked to severe drug reactions like Stevens-Johnson syndrome when patients take carbamazepine.
Pan et al. conducted a study on Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN), traditionally considered a T-cell-mediated disorder. They used next-generation sequencing to analyze the T cell receptor (TCR) repertoire and discovered a public αβTCR from the cytotoxic T cells of patients with carbamazepine-induced SJS/TEN. This public αβTCR was found to bind with carbamazepine (CBZ) and mediate an immune response, providing insights into the mechanisms driving severe cutaneous adverse reactions.
