Development of Fast Reverse Phase Liquid Chromatographic Method for Simultaneous Analysis of Betamethasone Dipropionate and Calcipotriol in Ointments and Bulk Drug

    In 2014, researchers developed a reverse phase isocratic liquid chromatographic method for analyzing betamethasone dipropionate and calcipotriol in ointments and bulk drugs. The method utilized a water and methanol mobile phase, a C-18 column, and UV detection at 265 nm, achieving retention times of 4.5 and 6.9 minutes for betamethasone dipropionate and calcipotriol, respectively. It demonstrated excellent linearity with correlation coefficients of r² = 1 for calcipotriol and r² = 0.999 for betamethasone dipropionate, and high recovery rates of 98.6% and 99.5%. The method was validated with relative standard deviations of 0.67% and 1.14% for the two drugs, respectively, and proved to be robust, cost-effective, and in compliance with ICH guidelines. The method showed less than 2% variation under altered conditions, and solution stability was confirmed with less than 1.5% variation over 24 hours. It was considered suitable for routine analysis of these drugs in pharmaceutical formulations.
