This study explored the roles of E2 enzymes in unconventional ubiquitination in Arabidopsis thaliana, focusing on UBC13 and UBC22. UBC13 was linked to Lys63 polyubiquitination, affecting root development and sugar signaling, with ARF6 overexpression partially rescuing root phenotypes in ubc13 mutants. UBC22 was identified as an E2 for Lys11-linked ubiquitination, crucial for seed setting and female gametophyte formation. UBC22 mutants exhibited varied vegetative development phenotypes, with some showing enhanced pathogen resistance. The findings advanced the understanding of E2 functions in plant ubiquitination, highlighting UBC13 and UBC22's roles in development and stress responses.
