    The study utilized site-directed mutagenesis to explore the role of specific amino acid residues in human steroid 5alpha-reductase types 1 (5AR1) and 2 (5AR2) in inhibitor/substrate binding. It was found that replacing residues 21-24 in 5AR2 with those from 5AR1 did not significantly affect binding characteristics, while replacing residues 26-29 in 5AR1 with those from 5AR2 decreased the Km and increased resistance to Finasteride. This suggested that residues 26-29 in 5AR1 are crucial for binding, unlike residues 21-24 in 5AR2. Further experiments showed that residues 15-17 in 5AR2 are involved in binding, whereas residues 18-20 are not. These findings provided insights into the specific roles of amino acid residues in the binding process of these enzymes.
