This study investigated the impact of hair pigmentation on the binding of pharmacologically interesting molecules using an in-vitro model. It examined the concentration and time-dependent uptake of foreign substances in immortalized cell lines simulating the hair shaft. The study found different binding properties in keratinocytes and melanocytes, with unpigmented keratinocytes reaching saturation of foreign substance uptake faster than pigmented melanocytes. In melanocytes, binding to melanin was significantly stronger than to cellular proteins. Primary melanocyte and keratinocyte cell populations isolated from human hair follicles and epidermis showed that epidermal melanocytes and outer root sheath melanocytes were suitable for in-vitro studies of foreign substance incorporation.
