Establishment of Tsc2-Deficient Rat Embryonic Stem Cells

    TLDR Tsc2-deficient stem cells can help understand and treat TSC-related tumors.
    The study established Tsc2-deficient embryonic stem cells (ESCs) from Eker rats, which maintained pluripotency and could differentiate into all three germ layers. Tsc2−/− ESCs showed abnormal mTORC1 pathway activation and distinct gene expression profiles compared to Tsc2+/+ and Tsc2+/- ESCs. These cells could form teratomas but caused embryonic lethality in chimeras. The research suggested that Tsc2 deficiency might lead to abnormal differentiation, potentially affecting hair development, and highlighted the use of these ESCs as models for studying Tsc2 mutation-related pathogenesis and identifying therapeutic targets.
