RY-023, an Inverse Agonist at Alpha5 GABAA Receptors: The Influence on Spatial Memory and Spontaneous Locomotor Activity

The study from 15 years ago investigated the effects of RY-023, a benzodiazepine site inverse agonist with 75-fold selectivity for the α5nY2 receptor subtypes, on spatial learning and locomotor activity in male Wistar rats. The study involved 7 rats per group, which received varying doses of RY-023 each day for 5 consecutive days. The results suggested that ligands with appreciable inverse agonist activity at GABAA receptors containing α5 subunits may enhance memory acquisition in early stages of learning, without discernible effects on general activity. However, these ligands appeared to be less effective in modulating the later stages of the learning process and likely exerted no delayed influences on memory recalling.
