This study investigates the roles of γδ T cells across different mouse tissues, revealing that these cells are not a homogeneous population but consist of unique site-adapted subsets. Through multimodal single-cell profiling and parabiosis experiments, the research identifies distinct cellular states linked to functions such as cytotoxicity, rapid interferon-γ production, and tissue repair, all displaying tissue residency characteristics. The study highlights the complexity of interleukin-17-producing γδ T cells in relation to tissue residency and finds that the transcriptional programs of tissue-resident γδ T cells are similar to those of CD8+ tissue-resident memory T cells. This research provides a detailed view of the heterogeneity and tissue residency programs of γδ T cells.
