From Primary MSC Culture of Adipose Tissue to Immortalized Cell Line Producing Cytokines for Potential Use in Regenerative Medicine Therapy or Immunotherapy

    The study developed an immortalized cell line, HATMSC1, from adipose tissue mesenchymal stem cells (MSCs) to produce cytokines for regenerative medicine and immunotherapy. HATMSC1 showed higher cytokine production than primary MSCs, with significant levels of factors like Angiogenin and MCP-1. The cell line's supernatant enhanced proliferation, adhesion, and angiogenesis in wound healing models, particularly benefiting fibroblasts and endothelial cells. Hypoxic conditions and stimuli like TNF-alpha increased cytokine production and altered cell phenotypes. The study involved 8 patients with nonhealing wounds, suggesting HATMSC1's potential in therapeutic applications due to its robust cytokine production and responsiveness to environmental stimuli.
