The study investigated the impact of the Wnt signaling pathway on the reprogramming of human somatic cells into induced pluripotent stem cells (iPSCs). It was found that activating Wnt/β-catenin signaling, particularly through Wnt3a or β-catenin overexpression, significantly enhanced the formation of pluripotent colonies. Conversely, inhibiting Wnt signaling reduced reprogramming efficiency. The research suggested that Wnt/β-catenin signaling was essential for iPS cell reprogramming and could provide a safer alternative to viral transduction methods, which posed oncogenic risks. The study also speculated on the potential of addressing disorders like focal dermal hypoplasia by restoring Wnt signaling pathways.
