Reconstitution of Human Skin In Vitro Using Readily Available Sources: Novel Studies of Tumor Promotion

TLDR A lab model of human skin was created to study skin tumor promoters without using actual human skin.
The study developed a standardized three-dimensional in vitro model of human skin to assay skin tumor promoters. This model was notable for its replication of human skin characteristics, the ability to seed with "initiated" keratinocytes, and control over the medium's calcium level. It combined methodologies from previous research and did not require actual human skin, making it reproducible in any lab with basic cell culture equipment. The model used human embryonic fibroblasts and rat tail collagen for the dermal equivalent, with keratinocytes from human hair follicles. Stratification was achieved by culturing epidermal cells at the air interface of a serum-free medium, and the model was validated through histological staining, electron microscopy, and immunostaining.

Oral minoxidil is effective for hair loss but can cause cardiovascular side effects, such as pericardial effusion, especially in those with genetic predispositions. Starting with low doses and monitoring cardiovascular health are advised to reduce risks.

community Minoxidil is NOT an Anti-androgen. No.

Minoxidil is not an anti-androgen; it is a growth stimulator and does not lower DHT like finasteride or dutasteride. Minoxidil can cause side effects like facial swelling and anxiety due to its vasodilator properties.

Minoxidil sulfate is more effective than regular minoxidil, especially for those with low sulfotransferase levels or scalp sensitivity, but it is unstable unless delivered in a liposomal format. Combining minoxidil with tretinoin can enhance effectiveness, and stopping minoxidil use can lead to rapid hair loss.

community AGA responded to steroid shots? Biopsy wrong? Please help!

A woman with AGA is using spironolactone, dutasteride, finasteride, oral minoxidil, bicalutamide, and anti-androgenic birth control but still experiences worsening hair loss. Steroid shots temporarily stop her hair shedding, leading her to question her biopsy results.
