Epidermal Stem Cells Manipulated by pDNA-VEGF165/CYD-PEI Nanoparticles Loaded Gelatin/β-TCP Matrix as a Therapeutic Agent and Gene Delivery Vehicle for Wound Healing

    The study from June 28, 2013, investigated a nonviral gene delivery system using a gelatin/β-TCP scaffold loaded with CYD-PEI/pDNA nanoparticles to enhance wound healing by transfecting epidermal stem cells (ESCs) with the VEGF165 gene. The in vitro results showed that this 3D transfection system had higher efficiency compared to conventional methods. In vivo experiments on rat skin wounds demonstrated that the transfected ESCs significantly improved wound closure, dermal collagen synthesis, hair follicle regeneration, and angiogenesis, with potential for scar inhibition. The study concluded that the gelatin/β-TCP scaffold loaded with CYD-PEI/pDNA nanoparticles is an effective therapeutic agent for wound healing, promoting skin regeneration through increased VEGF expression and angiogenesis.
