Construction and Activity Analysis of K14/K5 Promoter Vector Containing Luciferase Gene

    The study focused on constructing pGL3-K14 and pGL3-K5 promoter vectors by inserting K14 and K5 promoters into the pGL3-Basic vector. These promoters, known for directing tissue-specific expression in stratified epithelia, were analyzed using luciferase reporter assays and GFP expression tests. The results indicated that both promoters were expressed across all tested cell lines, with the K14 promoter showing higher activity specifically in skin cell lines compared to others.
