    This study investigates the role of perforin-mediated cytolysis in the development of Alopecia Areata (AA), an autoimmune disease causing hair loss. Researchers used C3H/HeJ mice deficient in perforin (PKO) and found that these mice could still develop AA spontaneously. In a skin-graft induction model, both wild type (WT) and PKO mice showed similar onset and progression of AA. However, in a CD8-induction model, CD8 T cells from PKO mice were less effective at inducing AA compared to those from WT mice. The findings suggest that perforin-mediated cytolysis is not essential for the autoimmune attack on hair follicles, indicating that other mechanisms of CD8 T cells contribute to AA.
