The study explored the role of TRPV1-mediated signaling in sebaceous gland biology using a human sebocyte cell culture model. It was found that TRPV1 is expressed in human skin sebaceous glands and SZ95 sebocytes, and that the TRPV1 agonist capsaicin affects lipid synthesis and cell proliferation in a dose-dependent manner. Low doses of capsaicin stimulated cell proliferation via TRPV1, while higher doses inhibited growth and induced cell death independently of TRPV1. Capsaicin also suppressed genes involved in lipid homeostasis and proinflammatory cytokines. These findings suggested that TRPV1 signaling played a significant role in sebocyte biology and could be a target for managing inflammatory sebaceous gland disorders like acne vulgaris.
